Happy #moleculemonday! Cannabidiol aka CBD has officially hit the fitness supplement market! CBD is a cannabinoid found in marijuana(both hemp and sativa) that is not psychoactive like THC, but does have many amazing properties for both healthy and disease-state people. For healthy, weight training people, CBD has many interesting applications including arthritis, pain relief, anxiety, inflammation(systemic), increases appetite, cardio health, sex drive, sleep trouble, and even may lower incidence of getting type 2 diabetes. CBD has also been shown to have many positive effects for treating cancer including inhibiting tumor growth, inducing apoptosis of cancer cells, preventing development of blood vessels for tumors, and the best part is it seems to ONLY AFFECT CANCER CELLS(in vitro), not healthy cells!! For me, I'm very excited about the application of CBD for inflammation and arthritis. I've had 3 surgeries on my left wrist(I'm left handed) which has affected my training, as well as I have a condition known as Barrett's esophagitis. I will be doing a pilot study on myself for the treatment of acid reflux with CBD. The endocannabinoid system in our bodies has so many interactions and crosstalk with many of our other systems, including appetite control, energy balance and metabolism, so the applications could be almost endless! The legal status of CBD is very gray to say the least. The gel caps in this pic are from hemp, not sativa, so it may be protected by the agricultural act of 2014. At the same time, CBD is currently a schedule 1 narcotic in federal law, while legal in some form in 29 states(I think). This can cause issues for transporting CBD from a legal state to a non-legal state. This post goes out to all the hardcore bodybuilders who use "supplements." Two of the major side effects of these "supps" is the unfavorable change in HDL/LDL and loss of Ca from bones. Both of these can affect calcification/hardening of arteries which can cause serious problems(ie heart attacks). The list of bodybuilders who have passed away or who had heart related conditions in too long to list. One supplement that should be a staple is Vitamin K2. K2 has been extensively studied and shown to reduce both calcification of arteries and death from heart disease. In a 7 year study showed a 57% reduction in death from heart disease. Another study showed a significant decrease in likeliness of heart attack and calcification. The dose studied was > 32.7mcg. I take 90mcg/d. K2 seems to interact with calcium-regulating proteins in bones to help spare Ca and prevent it from depositing on arteries. If you regularly take anabolics or prohormones, this is a supp definitely worth taking. As most of you guys know, thyroid hormone regulates the metabolism of proteins, fats, and carbs and affects body composition. It determines how "lean" you stay year around(we all have that one friend that eats way more than you and still has abs 24/7 @lawrenceballenger). GC-1 is a thyroid hormone receptor(TRbeta) agonist "thyromimetic" (mimics thyroid hormone without being a hormone) that shows some very promising results in the way of fat loss, body comp, ect. Basically, GC-1 acts similarly as T3(thyroid hormone) in the body, but doesn't seem to cause suppression of TSH(thyroid stimulating hormone) levels, a side effect of supplementing with T3/T4/T2. What the studies have shown so far is that GC-1 can increase basal metabolic rate(and fat burning) without causing tachycardia, a side effect from T3. When compared head to head, T3 caused a slightly greater reduction in body weight vs GC-1, but both caused significant increases in energy expenditure and fat loss, while preventing fat mass accumulation. Another benefit noticed with GC-1 was the sparing of bone mineral density, something that is reduced when taking T3. GC-1 recently has been shown to help turn white fat into brown fat. Brown fat is high in UCP-1, a protein that upregulates fat burning via uncoupling. The latest study in mice showed a 50% loss in fat mass in just 2 weeks, as well as a 6-fold improvement in insulin sensitivity. GC-1 (Sobetirome) is currently in Phase 1a clinical trials for its cholesterol lowering effects. More research is needed, but this definitely looks promising and should hit the bodybuilding circuits. TB500 is a peptide fraction of the protein Thymosin beta4, which is released in times of healing and inflammation. TB500 main mechanism is its ability to regulate Actin-a cell building protein that is present in every cell. By upregulating Actin, your body can now increase cell proliferation cell migration. It also helps promote wound healing, and has been shown to build new blood vessels/endothelial factors, increase keratinocyte migration and increased collagen deposition and muscle fiber. It has has been shown to downregulate inflammatory chemokines(white blood cell attraction) and has even been shown to promote cell survival. There are not a lot of human studies on this peptide at the moment, but all the animal data seems promising, not to mention the tons of anecdotal evidence for healing. There have even been reports of men regrowing lost hair and gray hair turning darker. There is an animal study showing hair follicle development so this is plausible. As for the dosing protocol, it's seems to work best by using a loading phase, 4-6 weeks at 4-8mg per week split up into 2mg subQ injections(TB500 is systemic). This can be followed by a maintenance phase of 2-4mg every 2 weeks, again in 2mg doses. It really depends on the severity of the injury and bodyweight to determine an exact protocol for you. It seems to work best on ligament and tendon injuries, but there is evidence for repairing heart damage following a heart attack. This is best stacked with BPC-157 at 500-1000mcg a day (on an empty stomach). Worth noting, there have been studies showing increased cancer cell proliferation with TB500, but only in pre existing subjects. Happy #moleculemonday! With DMAA and DMHA all but gone, Senegalia extract, an acacia species, has popped up as a new replacement. This picture is from a paper that identified with GC-MS all the amine/alkaloids present in this plant. As you can see, there are some extremely powerful stimulants, including naturally occurring amphetamines, nicotine, mescaline, and various phenylethylamine derivatives. I have not tried this in a product yet, but I'm interested to see what compounds were extracted and used, and how strong it is as a possible new stimulant. Here's a #TBT that might open your eyes to the evolution of the sport. This is a picture of the winners from the 2009 NPC Nationals for bikini(left) and figure(right). By today's standards, the bikini winner wouldn't even place at a local show and the figure winner would probably place for bikini. For you ladies who just started competing, this is what was envisioned when the bikini class was first started. Interesting to see how the sport will always get bigger, harder, leaner, ect. I was lucky enough to get some beta samples of @optimresearchinfo new product 11-oxo acetate. If you remember, 11-oxo AKA adrenosterone was a 11beta hydroxysteroid dehydrogenase inhibitor, the enzyme that converts cortisone to cortisol, was sold as an OTC supplement brought to the market by Patrick Arnold. The ketone(oxo group) on carbon 11 accepts a hydride(a hydrogen atom with 2 electrons) instead of cortisone. This supplement was great for fat loss due to reducing cortisol. It's also a weak androgen and in higher doses can increase lean muscle. Im very excited about this product and its potential for fat loss. I will keep you posted about my experience with it. The acetate ester will require EOD injections but you can also do small subcutaneous injections(0.3mL max). I would recommend weekly doses of 300-500mg/week for fat loss. Just ran across this 2014 study showing that a 50/50 combination of citrulline/arginine mix elevated plasma arginine levels and blood nitrate/nitrite levels higher than either compound alone at 2x the dose, in both rats and rabbits. Interestingly, the Citrulline/Arg blend was only more effective for the initial peak, which lasted for 60min. After that, levels of NOx and Arg were about the same in all 3 groups. The initial increase was about 35% higher in NOx and Arg levels, which is pretty significant. Previously studies have shown that Arginine can elevate NO levels only in very high amounts, 9g and above. Worth noting that Arg has a higher affinity to the enzyme arginase vs NO synthase, and arginase breaks down Arg into orthinitine. Researchers hypothesized that citrulline acts as an allosteric inhibitor(binds to the enzyme but not the active site, essentially changing the shape of the enzyme) of Arginase, which supports the data of higher Okasan arginine levels vs just arginine. The problem is L-carnitine isn't well absorbed orally, and even injectable carnitine doesn't really elevate cellular concentrations. Enter insulin. Carnitine uses an active transporter called OCTN2 to bring it across cell membranes. Insulin activates the Na/K ATPase pump which facilitates pumping Na+ ions into cells and K+ out. OCTN2 has an affinity for Na+ and it can bind to Na and then carnitine and be carried into the cell via the Na/K ATPase pump. Researchers showed that spiking insulin via glucose while taking carnitine was able to increase muscle carnitine levels by 13% vs 0.1% without. Of course, you can stimulate insulin without carbs via injecting a small amount, 1-2IU with your carnitine to ensure maximum muscle carnitine levels. GSH is the key compound in the liver responsible for metabolizing tons of molecules, including alcohol and methylated androgens. The thiol(SH) group is the most important part of the molecule, as it is one of the only functional groups that can interact with the major free radicals/oxidants that your body naturally produced. These oxidants are responsible for cell damage, as well as DNA and mitochondrial damage which can cause significant health issues and is the root cause of aging. GSH levels decline significantly with age. The main problem with GSH is that it has bad oral bioavailability and cannot enter cells as the tripeptide. As promised, a post about BPC 157 and the proper dosing protocol. BPC is a peptide comprised of 15 amino acids originally developed as an anti ulcer agent and IBS treatment. In fact, the peptide is a fragment of a naturally-occurring gastric protein. It's gained a lot of popularity though for its anti-inflammatory properties as a treatment for arthritis. One of its main mechanism of actions is ability to influence several growth factors involved in the production of new blood vessels. In mice studies, BPC has been found to increase rate of growth of fibroblasts(tendon cells) as well as improve collagen reformation rates, which may explain its healing benefits users are experiencing. There are also studies showing increased bone and tendon healing, via increases growth factor expression. As far as dosing, benefits can be seen from taking 250mcg-1000mcg a day for up to 4 weeks. Despite what people on forums say, the research suggests BPC works systemically. Some people insist on injecting at the site of injury, but I do not feel it's needed, seeing as the drug can be injected subcutaneously or taken orally. So, I'm not a fan of protein powders and prefer to get my protein from while food sources, but I understand that they can be helpful both monetarily and conveniently. I got burnt out on them in grad school, but they definitely helped me gain size. To me, when comparing the top tier proteins, the most important part of a protein powder is taste and Isofract tastes insanely good. I prefer a thicker, richer protein shake vs isopure or gold standard which are watery IMO. I tried the chocolate and it was very rich, which surprised me Bc it's naturally flavored with stevia and monk fruit. If you're looking for a top quality protein that tastes really good, this is my recommendation. On a side note- I like this company #myoblox a lot, specifically Bc of their branding(photo doesn't do the label justice). The owner is an artist and he does these awesome limited flavor releases vis-a-vis limited sneaker drops, which to me is pretty badass marketing. Is trestolone the new testosterone? Trestolone was first synthesized as a male birth control option, when researchers were looking for a safer alternative to test. Test converts to DHT, which can have deleterious effects on your prostate in the long term. Trestolone, due to its alpha-methyl group on carbon 7, cannot convert to DHT via 5a-reductase. This methyl group also prevents it from binding to sex hormone binding globulin, the protein that renders test/estrogen inactive. This means sex drive is very high while on cycle, something 19-nortestosterone derivatives are not known for. In a study in healthy males comparing trestolone to test, all health biomarkers including HDL/LDL, PSA, total triglycerides, and hematocrit levels all were less affected than testosterone. Being a male contraceptive candidate, trestolone significantly lowered both LH and FSH, 90% and 91%, respectively in 4 weeks. Levels returned to baseline post cycle. The other major side effect is trestolone can readily convert to a strong estrogen, 7a-methyl-estradiol which also cannot bind to SHBG. An aromatase inhibitor is strongly recommended. IMO, trestolone is significantly more anabolic than testosterone, mg for mg. I had great success using 50mg of the acetate EOD and am awaiting to try the decanoate ester, in the 300-400mg/week range. One of the most important things you should do when taking any prescription drug for bodybuilding purpose is READ THE PAMPHLET on how the drug works. It amazes me the stories I hear about competitors taking something like a potassium-sparing diuretic, then cutting sodium, water and then ADDING potassium! This is a recipe for disaster and you will never be able to carb up. Your body needs both sodium and water to store glycogen and if you are messing with sodium/potassium levels in your body, it will affect the Na/K pumps that control cellular osmolality and muscle contractions. You would know this if you read the pamphlet on how the drug works in patients who need it. This is just one example, but it's always a good idea to read how people prescribed these drugs take them and why the doses prescribed the way they are. I cannot stress this enough. I will guarantee that 75%+ of "coaches" do not do this. Educate yourself! These are dangerous drugs that you are using in a way for which they are not intended. Read the pamphlet! This was a fan request and I always try to give the people what they want lol.  Cabergoline(caber) is a dopamine D2 receptor agonist that directly inhibits prolactin. As you may know, gynecomastia can be caused by both estrogen and progesterone(indirectly), which can be elevated by exogenous androgens.  Progesterone can stimulate prolactin serum levels which can increase mammary gland size. Caber works by directly inhibiting prolactin secretion in the pituitary's lactotroph cells. Interestingly, prolactin has a very similar structure to GH, and studies have shown the use of caber to decrease GH levels. Another interesting phenomenon observed in male caber users is multiple orgasms, due to its reductive effect on the refractory period(time between ejaculations). As far as side effects, caber is the safest option for hyperprolactinemia and bc of its low effective dose(0.25-1mg 2x a week) most common side effects being gastric related and loss of appetite. Overall, caber is very effective at reducing prolactin buildup caused by certain androgens like nandrolone and trenbolone.
